چکیده

D-(-)-ribose was subjected to oxidation with equimolar amounts of H20 2/Fe2+ and 13-hydroperoxy-9-c/s-l l-fra/75-octadecadienoic acid/Fe2+. Smaller carbohydrates, carbonyl compounds, hydroxy acids and polyols were produced. Identification of carbonyl compounds was achieved by trapping with pentafluorobenzylhydroxylamine hydrochloride (PFBHA-HC1). All products were identified by GC/MS after derivatization with N-methyl-N-trimethylsilyltrifluoroacetamide (MSTFA). The reactions were repeated with D-[13Ci]-ribose in order to reveal the mechanism of oxidation. A possible reaction pathway is outlined starting by re moval of hydrogens from C-H bonds.

Reduction of alkyl hydroperoxides to alcohols: role of rubredoxin, an electron carrier in the bacterial hydroxylation of hydrocarbons.

Alkyl hydroperoxides are reduced to alcohols in the presence of DPNH and homogeneous rubredoxin and rubredoxin-DPN reductase from Pseudomonas oleovorans. The reaction does not occur when TPNH is substituted for DPNH or when rubredoxins from anaerobic bacteria are substituted for the P. oleovorans nonheme iron protein. The reaction appears to have broad substrate specificity; 1-octyl, 2-octyl, c...

Mechanism of the antioxidant to pro-oxidant switch in the behavior of dehydroascorbate during LDL oxidation by copper(II) ions.

Oxidised low density lipoprotein (LDL) may be involved in the pathogenesis of atherosclerosis. We have therefore investigated the mechanisms underlying the antioxidant/pro-oxidant behavior of dehydroascorbate, the oxidation product of ascorbic acid, toward LDL incubated with Cu(2+) ions. By monitoring lipid peroxidation through the formation of conjugated dienes and lipid hydroperoxides, we sho...

Low-density lipoprotein is the major carrier of lipid hydroperoxides in plasma. Relevance to determination of total plasma lipid hydroperoxide concentrations.

High-density lipoprotein (HDL) has been proposed as the principal carrier of hydroperoxides in plasma, based upon data gathered with an HPLC-chemiluminescence technique. To test this hypothesis we have measured total lipid hydroperoxides in native plasma using the ferrous oxidation in Xylenol Orange (FOX) assay and then fractionated plasma into very-low-density lipoprotein (LDL) and HDL fractio...

DHA Hydroperoxides as a Potential Inducer of Neuronal Cell Death: a Mitochondrial Dysfunction-Mediated Pathway

During the lipid peroxidation reaction, lipid hydroperoxides are formed as primary products. Several lines of evidence suggest that lipid hydroperoxides can trigger cell death in many cell types, including neurons. In a screening of lipid hydroperoxides which can induce toxicity in neuronal cells, we found docosahexaenoic acid hydroperoxides (DHA-OOH) induced much severe levels of reactive oxyg...

Annihilation of Leishmania by daylight responsive ZnO nanoparticles: a temporal relationship of reactive oxygen species-induced lipid and protein oxidation

Lipid and protein oxidation are well-known manifestations of free radical activity and oxidative stress. The current study investigated extermination of Leishmania tropica promastigotes induced by lipid and protein oxidation with reactive oxygen species produced by PEGylated metal-based nanoparticles. The synthesized photodynamic therapy-based doped and nondoped zinc oxide nanoparticles were ac...
